  • Descrição: Objetivo: Chemical modifications of LDL elicit specific humoral response against new apo B epitopes and may contribute to inflammatory response and atherogenesis. Serum detection of these specifics surrogate markers in CAD patients are under current investigation by several groups. Our aims were analyse the presence of antibodies aginst defined and specific components of oxLDL in serum samples obtained from CAD patients by ELISA assay. Métodos e Resultados: Human purified LDL was oxidized in vitro (20 uM CuSO4, 18h). The resulting preparation was separated by Superdex-200 column (Amersham-Pharmacia-Biosciences, Sweden) and the eluate was monitored with a diode-array detector (Shimadzu, Japan). The peak fractions detected at 280 nm were then tested in ELISA. In these assays, serum from healthy individuals were compared to patients with angiographically confirmed coronary artery disease (CAD) The size-exclusion chromatographic profile showed 4 peaks. Ten fractions from these peaks were tested in ELISA for reactivity to antibodies in healthy or CAD sera. Statistical analysis of these fractions allowed us to detect significant difference among 6 of them. Three fractions containing high mass oxLDL components exhibited high reactivity with sera from healthy individuals as compared with those of CAD individuals. ) Those containing low mass components exhibited the inverse property. Conclusões: A number of antigens with different properties can be genereted by
    means of copper-induced modification of LDL and chromatographic separation is a powerfull method to separate and characterize potential oxLDL antigens. The reactivity of the serum antibodies of CAD individuals against purified oxLDL antigens showed some specific antigens that can help us in the future to improve the diagnosis and treatment of atherosclerosis
