  • Descrição: Despite the central role of FtsH proteases in cellular processes, the expression of these proteins are poorly understood. In this work we propose to study the expression and subcellular localization of FtsH-p1 protease from Arabidopsis thaliana. Material e/ou métodos: Using the genome sequence of the Arabidopsis thaliana available at NCBI, we amplified by PCR the promoter region of the FtsH-p1 as well as a 1365 pb region in located within a previous orf. These sequences were cloned in the vector pCAMBIA-1301. To determine the subcellular localization of the FtsH-p1, a transient expression system in onion cells was carried out. Then, the 5 region encoding the putative targeting sequences followed by two transmembrane segments were fused in frame to the Green Fluorescent Protein (GFP) gene in the pCAMBIA 1302 vector, under the control of the 35S promoter. After bombardment, the subcellular location of the GFP was visualized on a fluorescent microscope. Resultados: The sequences related to promoter region of the FtsH-p1 gene was successfully amplified by PCR and cloned into the pCAMBIA-1301 vector in order to drive the expression of the gus reporter gene in transgenic Arabidopsis plants. GFP fused to the N-terminal region of sugarcane FtsHs-p1 was specifically targeted to the plastids. Conclusões: The study of the protein expression is an important method in the agreement and characterization of genes with potential of use in genetic engineering
