  • Descrição: Objetivo: The ability of the early environment to program the hypothalamo-pituitary-adrenal axis (HPA) promoting neuroendocrinological and immune alterations in adult life has been well documented. We have investigated whether prenatal administration of LPS affects the allergen-induced airway leukocyte migration in adult offspring, and the modulatory role of NO in this inflammatory response. Métodos e Resultados: Pregnant Wistar rats were divided in 5 groups: 1) Control (non-manipulated during pregnancy); 2) Saline (i.p. administration of saline in 17th day of pregnancy); 3) LPS (i.p administration of LPS at 7 _g/Kg in 17th day of pregnancy); 4) Aminoguanidine (administration of aminoguanidine in drinking water from the 13th day of pregnancy to birth); 5) AG+LPS. In adult life, the male offsprings were sensitized with ovalbumin (OVA; 200 _g, sc, in AlOH3). Fourteen days after OVA sensitization, animals were challenged intratracheally with OVA (0.4 ml, 0.25%), and 48h thereafter the bronchoalveolar lavage (BAL) fluid was examined regarding the leukocyte counts. Prenatal LPS administration reduced significantly (p<0.01) the eosinophil influx (0.82±0.16 x 106 cells/BAL) as compared with saline (2.04±0.37 x106 cells/BAL). Pretreatment with aminoguanidine restored the eosinophil influx in LPS-injected rats (2.32±0.24 x106 cells/BAL). When given alone, aminoguanidine had no significant effect in the OVA-induced cell influx. Conclusões: Prenatal LPS
    administration decreases the organism capacity to respond properly to a pulmonary allergic stimulus in adult life. Here we demonstrated that NO plays a critical role in this process, since the iNOS inhibitor aminoguanidine was able to abolish this phenomenon.
