  • Descrição: Introduction and Objectives: B-1 cells are classified as B-lymphocytes because they rearrange immunoglobulin genes, synthesize IgM and IgD and express characteristic lymphocyte surface molecules (B220 and CD19). However, these cells also express macrophage surface markers (CD11b and F4/80) and differentiate in vitro into a mononuclear phagocyte, provisionally named lymphophage (Popi et al, submitted). Although they have some characteristics of macrophages, nitric oxide (NO) production by these cells is different. Results from our laboratory suggest that B-1 cells produce NO constitutively. Herein we investigated NO production by B-1 cells and characterized the NO synthase responsible for their NO production. Methods and Results: B-1 cells and lymphophages were cultivated as described by Popi et al (submitted). Macrophages were obtained from bone marrow cells cultures cultivated in LCCM medium. Nitric oxide release in the culture medium and intracellular NO expression were analyzed in cultures described above, stimulated or not with 10ng/ml of LPS. Results show that NO production of B-1 cells increase up to five days cultures. Lymphophage, however, decrease the intracellular production. LPS do not stimulate NO production in both B-1 derived cultures, although it increases NO production by macrophages. Western blotting analysis of NO synthase isoforms showed the presence of iNOS even in B-1 cells and their phagocyte derived cells without stimuli. The
    expression of constitutive NOS by these cells is under investigation. Conclusion: These results support evidence of a spontaneous NO production by B-1 cells and that these cells have a constitutive expression of iNOS.
