  • Descrição: Objetivo: The occurrence of DNA fragmentation in lymphocytes obtained from diabetic patients and alloxan-induced diabetic rats was investigated. Métodos e Resultados: Blood human lymphocytes and rat mesenteric lymph node lymphocytes were analysed for DNA fragmentation by using flow cytometry and agarose gel and for chromatin condensation by Hoescht 33342 staining. Immediately after being obtained, the proportion of lymphocytes with fragmented DNA was 76% greater in diabetic patients compared with cells from healthy individuals. The occurrence of DNA fragmentation remained 50% higher in lymphocytes from diabetic patients even after 48 h in culture. Similar results were obtained in lymphocytes from alloxan-induced diabetic rats. The involvement of high glucose levels and/or low insulinemia for the occurrence of apoptosis in lymphocytes was also examined. There was no relationship between plasma glucose levels and the proportion of lymphocytes in apoptosis. Also, high glucose concentrations did not cause DNA fragmentation in human and rat lymphocytes cultured for 24 and 48 hours. However, insulin treatment markedly reduced the proportion of lymphocytes with fragmented DNA in alloxan-induced diabetic rats. Conclusões: Diabetic states lead to lymphocytes death that is prevented by insulin administration
