  • Descrição: MYGALIN was purified by RP-HPLC from the hemocytes of unchallenged tarantula spider Acanthoscurria gomesiana showing activity against Escherichia coli. Such antibacterial activity is inhibited by catalase, which suggests that its mode of action involves production of hydrogen peroxide (H2O2). The molecular mass of MYGALIN determined by ESI-MS is 417Da. Another low molecular-weight antibacterial compound with catalase-inhibitable activity, N-ß-Alanyl-5-S-glutathionyl-3, 4-dihydroxyphenylalanine (5-S-GAD), has been purified from Sarcophaga peregrina and fully characterized (J. Biol. Chem. 271(23): 13573-7, 1996). Besides the antibacterial activity, 5-S-GAD showed phosphorilation inhibition in protein tyrosine kinases, apoptosis induction in monocyte-macrophage lineage cells and selective citotoxity to certain tumour cell lines. The inhibition of the antibacterial activities of MYGALIN and 5-S-GAD by catalase indicated us that these molecules could be structurally related. With the aim of investigating this hypothesis we carried out tandem mass spectrometry experiments (ESI-MS/MS). This partial structure analysis revealed differences with 5-S-GAD. Indeed, the results obtained suggest that MYGALIN consists of two dopamine molecules plus one molecule of 145 Da not related to natural amino acids. After concluding the full elucidation of MYGALIN structure we intend to explore the possibility of synthesising it enzimatically and study its unusual mechanism of action
