  • Descrição: Random Amplified Polymorphic DNA (RAPDs) analysis was used to evaluate the genetic diversity and variability present in 41 olive cultivars grown in Trás-os-Montes Region. Eleven RAPD primers were used. After agarose gels analysis, RAPDs banding patterns were transformed into binary data of presence-absence and matrices were processed with NTSYS-PC. Genetic distances between all pair wise combinations of the cultivars were calculated using Jaccard’s coefficient of similarity and dendrograms were constructed by the UPGMA method. All primers produced polymorphic bands. A total of 114 reproducible RAPD bands were observed, of which 100 were polymorphic, accounting for a high percentage (86%) of polymorphism. Five unique bands, able to be converted into Sequence Characterized Amplified Region (SCAR) markers, were obtained. Dendrogram similarity relationships revealed some clustering according to the olive cultivars geographical origin.
