  • Descrição: A bidirectional selective process to carry out a selective breeding of mice according to the presentation of a strong (AIRmax) and weak (AIRmin) acute inflammatory response was started from a foundation population constituted by the intercrossing of eight inbred strains (three of them are C5 deficient) for 36 generations. Considering that several mediators are derived from complement activation, our aim was to investigate if the intensity of the inflammatory response observed in AIRmax and AIRmin could be attributed to differences in the functionality of this system. Measuring the AP50 in each serum we observed significant differences (p<0.05) between sera from male and female animals in the two lines. Male and female AIRmax presented respectively 30U/muL and 44U/muL. Male and female AIRmin presented respectively 25U/muL and 47U/muL. When we considered all together no significant difference was observed between the two selected lines. Since the susceptibility to S. typhimurium is dependent on the natural-resistance-associated-macrophage protein (Nramp-1) gene and the resistant allele (r) is more frequent in AIRmax mice whereas the susceptible allele (s) is in AIRmin, we decided to analyse the alternative pathway in the sera from AIRmax and AIRmin homozygous for each allele of Nramp-1 gene. Surprisingly, no hemolytic activity was found in any serum from AIRmax r/r mice and no C5 was detected by immunodiffusion against anti-C5. However,
    serum from AIRmax s/s, AIRmin r/r and AIRmin s/s presente normal hemolytic activity and no significant difference between sera from each subline. In this manner, we have created a genetically selected group of outbred mice which have a strong acute inflammatory response, with a genotype for resistance to S. thyphimurium and lack of the component C5
