  • Descrição: Norwalk-like viruses (NLV) are a major cause of gastroenteritis in older children and adults and are associated with sporadic gastroenteritis and epidemic disease in families, schools, other institutional settings, and community wide outbreaks caused by contaminated water and food. NLVs are small, single-stranded positive sense RNA viruses, recently designated as a separate genus in the Caliciviridae family, based on phylogenetic analyses and genomic organization. These viruses have been divided into 2 genetically and antigenetically distinct genogroups: genogroup I (GI) and genogroup II (GII). The objective of this study was the identification, molecular characterization and phylogenetic classification of Norwalk-like strains in 188 stools samples from São Paulo city, Brazil, collected from children up to two years old with gastroenteritis, during a 5 years period (1995-1999). These samples were previously tested for rotavirus and adenovirus and were found negative for these viruses. The identification and molecular characterization were performed by reverse transcription-polymerase chain reaction (RT-PCR), with specific primers for GII of Norwalk virus. The GII positive samples were then submitted to a sequencing reaction for the 81bp segment of the viral polymerase gene. The sequence obtained was compared with sequences of NLV samples in GenBank. Preliminary results of 34 samples analyzed by RT-PCR, showed 6 (17,6%) samples positive for NLV of genogroup II.
    Comparison of nucleotide sequence 4 samples with NLVs sequences showed the following identities: two samples showed 86,4% identity with Bristol virus strain (BV) and two showed 98,8% and 97,5% identity with the 95/96-US strain. This is the first NLVs strain characterization by nucleotide sequencing in sporadic gastroenteritis in São Paulo city, Brazil
