  • Descrição: The aquatic bacterium Caulobacter crescentus is found in most humid habitats, from soil to marine water, surviving in environments that are poor in organic matter. The complete genome sequence of this bacterium was determined recently, but its response systems to environmental conditions are still poorly understood. We have analyzed the alteration in gene expression induced by cold shock in C. crescentus. The temperature range in which Caulobacter can grow was determined by analyzing growth curves and viability of cultures growing at 0 C, 5 C, 10 C, 15 C, and 30 C. Experiments of pulse labelling of proteins with 35S-methionine showed that at 10 C there is increased expression of a specific set of cold shock proteins (CSPs), including several of small MW. A detailed analysis of this differential expression was performed by two-dimensional gel electrophoresis of proteins pulse-labeled with 35S-methionine at 10 C during sequential time points. It was observed that the peak of expression of most of the CSPs was after 1 hour of incubation, and at least 50 proteins showed an increase in expression after this time. Three genes encoding homologs to E. coli CspA present in the C. crescentus genome were cloned. The promoter regions of each of these genes were cloned in a lacZ reporter vector and expression was analyzed by both beta-galactosidase activity assay and immunoprecipitation of 35S-labeled beta-galactosidase
