  • Descrição: Introduction and Objectives. Transcription factors T-bet and GATA-3 are important elements for the differentiation of CD4+ T lymphocytes into the functional phenotypes Th1 and Th2. While T-bet is necessary for the acquisition of Th1 phenotype and is of exclusive expression by these cells, GATA-3 is essential for the establishment and maintenance of the Th2 phenotype. In our experiments we intended to test whether the above cited transcription factors alone are sufficient to inducing epigenetic modifications on the loci of the cytokines IFN-g and IL-4, signature cytokines of Th1 and Th2 phenotypes, respectively. Methods and Results. By155.16 T cell lines were transduced with Tbet and gata3 carrying vectors (pMIG), sorted for GFP production and assayed for Hypersensitive Sites to DNaseI digestion (HSS) using radioactive probes specific for ifng and il4. Over-expression of the transcription factors was checked by western-blot analysis. Control, T-bet- or GATA-3-overexpressing cells did not differ in their hypersensitive sites distribution. Intracellular cytokine staining followed by flow cytometric analysis showed no increase in the levels of IFN-g and IL-4 by cells over-expressing the transcription factors when compared to controls. Conclusion. These results suggest a poor influence of the over-expressed transcription factors on the analyzed gene regions. Our data reinforce the notion that these factors are necessary but not
    sufficient for initiating the process of chromatin remodeling of il4 and ifng genes.
