  • Descrição: Objetivo: We demonstrated that losartan corrects the impaired endothelium-dependent vasodilation in mesenteric arterioles more easily in female than in male spontaneously hypertensive rats (SHR), (J Hypertension, 22:S176, 2004). Our objective was to investigate the role of oxygen reactive species (ROS), nitric oxide synthase (NOS) activity and angiotensin AT1 receptor expression on this difference. Métodos e Resultados: Male and female SHR (14-16 weeks) were divided into 03 groups (3-7 animals each) treated or not with losartan for 24 hours or 15 days (15mg/kg/day). After anesthesia (chloral hydrate, 500mg/kg, s.c.), the mesentery was exposed for microscopic observation and ROS generation was studied using the hydroethidine method. NOS activity in the mesentery vascular bed was evaluated using radiolabelled L-[3H]arginine conversion to L-[3H]citrulline (pmol/mg.min). mRNA and protein expressions for AT1-receptor in the mesentery bed were evaluated by RT-PCR and Westernblotting method, respectively. Losartan treatment for 24 h did not diminish ROS (in % ethidium bromide positive nuclei) in male (19±1.5% untreated vs 17.8±1.9% 24h) or female (11.4±1.5% untreated vs 14.9±2.0% 24h) SHR. Chronic or acute losartan treatment did not alter NOS activity in male (5.6±0.8 untreated vs 7.0±0.3 24h vs 3.9±0.4 15d) or female (5.9±0.8 untreated vs 6.2±0.3 24h vs 4.3±0.8 15d) SHR. Gene or protein AT1-receptor expressions were not modified by losartan treatment either in male or
    female SHR. Conclusões: Thus, differences in ROS generation, NOS activity or angiotensin AT1 receptor expression could not account for the differences in the responses of male and female SHR to losartan treatment.
