  • Descrição: Objetivo: Compared to men pre-menopausal women have less severe hypertension (H), but this protection is lost after menopause, with a rapid rise in pressure levels and consequent development of ventricular hypertrophy. We sought to investigate in females the effects of H, training (T) and ovariectomy on cardiac hypertrophy and possible regulatory signaling pathway involved. Métodos e Resultados: Intact (I) and ovariectomized (O) female SHR were submitted to T (treadmill, 50% VO2 peak for 3 months) or kept sedentary (S). Groups of WKYS and WKYT were used as controls. Left ventricles ( LV ) were removed, weighted and rapidly homogenized to extract total RNA for RT-PCR assay. LV weight was equally increased in both ISHR groups (+19% vs 585±13 and 622±15 mg in WKYS and WKYT), with a further increase in OSHRS and OSHRT (+7% vs respective ISHR, p<0.05). Only slight increases in LV mass were observed after T in all groups. CDK4 kinase expression, a pathological marker of hypertrophy was increased in ISHRS (1,09±0,21 vs 0.86±0.10 AU in WKYS, p<0.05), with a further 52% increase in OSHRS, but normalized by T (ISHRT=0,55±0,14, OSHRT=0.93±0.41 AU, p<0.05). T increased AKT expression (factor involved in physiological hypertrophy) in WKY and ISHR groups (+30% and +23%) but not in OSHRT; on the other hand, PPAR. (transcription factor associated with myocyte fatty acid metabolism) was increased by T only in OSHR (+78%). No Hinduced changes were observed for AKT and PPAR_
    expressions. Conclusões: Similar LV mass and CDK4 expression changes observed in SHR groups point out this factor as a good index for H-induced cardiac hypertrophy. Data also indicate that T-induced suppression of CDK4 signaling pathway is not accompanied by regression of LV hypertrophy in females. PPAR. and AKT expression are associated with small non-significant T-induced increases in LV mass (eccentric remodeling) in females exercised on treadmill
