Experiments were conducted to determine whether a radiotoxin is released not only when the hematopoietic system is irradiated, but also when any organ, tissue, or cell is so treated. Also considered was the question whether the toxic substances thus released are of the same, or varying, intensity and influence, depending upon their origin. A total of 153 rats were partiaily hepatectomized after which the regenerative capacity of the liver was tested following injection of various types of x-irradiated cell suspensions (liver, muscle, erythrocytes, and leukocytes) and whole blood. Irradiation was done in vitro at 180 kv, 15 ma, 0.5 mm Cu filter, and a rate of 67.6 r/min. The injected liver tissue per animal amounted to 0.15 to 0.20 g; muscle tissue, 0.10 to 0.15 g, was irradiated after being homogenized in saline and injected in a 1-ml suspension. Two ml whole blood, mixed with sodium citrate before irradiation, was injected. Blood cell suspensions (0.5 ml) were also irradiated and injected, the red cell suspension containing approximates 10,040,000 cells/mm/sup 3/ and the white cell suspension approximates 50,600 cells/mm/sup 3/. Various groups of anlmals received both irradiated and nonirradiated suspensions. Results show that irradiated liver, muscle, red cell suspension, and irradiated blood hindered iiver regeneration slightly. The results suggest that each of these irradiated tissues or cells releases radiotoxin. The irradiated embryo liver and leukocyte suspension, however, did not decrease the eight of the liver regenerating. The radiotoxin may represent catabolites or pathologic metabolites from cells having undergone radiation damage. These accumulated metabolites may then impair function of the organism. (BBB)
